酵母では、ORCは接合型(英語版)に関する遺伝子座Hidden MAT Left(HML)とHidden MAT Right(HMR)のサイレンシングの確立にも関与している[5][6][7]。ORCは、サイレンシングタンパク質Sir1をHMLとHMRのサイレンサーへリクルートすることによって、転写的にサイレンシングされたクロマチンの組み立てに関与する[7][17][18]。
